I see no reason why it wouldn't, it being a PC running Windows after all.

Just make sure that you initialize the new HDD as GPT (Globally Unique Identifier Partition Table) and not MBR (Master Boot Record):
    • MBR limits the max. size to 2TB no matter how big the drive is
    • GPT, on the other hand, "limits" the max. size to 9 400 000 000 TB. (Good luck finding a drive and having enough money to actually buy it, if you do find it!)
    More info HERE.

I believe your model is one of the Predator PO3-620 series. As such there are three SATA ports on the motherboard. Usually one is used by the ODD and the other two are available for HDDs or SSDs. There are two SATA power connectors on the MB, each of which can drive up to two drives. You may have to purchase a second cable for that if your machine didn't come with an extra in the box. The Acer part number for the power cable is: 50.E1ZD1.002. You will also need a standard SATA data cable to run between the MB and the drive. The HDD cage in your machine has room for two drives, so you should be good to go there.
