Ok.

Have you any Realtek icon on control panel or any tray icon?

if so:
Open the Control Panel, go to Realtek HD Audio Manager and if it has been updated you should see a little wrench in a box in the bottom right corner. Click this and inside there check the box next to "Disable front panel jack detection"

Ok.

we have 3 last things to check before ending with an hardware issue.

1) Open your Device manager, click on "sound, video etc etc" then double click on "realtek high definition sound"

on this windows check if "This device is working properly." message appears.

2) You can try to download from the Acer website the latest drivers available for your system, uninstall from the "program and feautures" the Realtek audio drivers, reboot and then install the downloaded package.

http://us.acer.com/ac/en/US/content/drivers/3297;-;Aspire%20Z3100

3) do a fresh installation of Windows

after 1 and 2, i can presume that something wrong happened on your motherboard or speaker connection, point 3 is the latest to check, just to be 100% sure that is an hardware related issue.

Re: my speakers wont work...solution? aspire z3100

No need to uninstall all but you will use Acer erecovery to re-install your OS, you will loose all your personal data.

by the way, you need to go to Device manager, no sound proprerties:

Open Device Manager by clicking the Start button, clicking Control Panel, clicking System and Security, and then, under System, clicking Device Manager, the open "sound" tree, double click on Realtek audio and check if driver is working.

If you're prompted for an administrator password or confirmation, type the password or provide confirmation.

Re: my speakers wont work...solution? aspire z3100

I had the same issue with my pc. I found out that some how my bluetooth had caused my internal speaskers to stop working when I tried to pair my phone. I had to remove the pairing of my phone and re load the blue tooth driver and that corrected the problem. Good luck
